It will be their first role together since playing Scott and Charlene in the Aussie soap.
Kylie, 54, and Jason, also 54, have already shot the episode, which will include nods to their famous pop career.
They topped the charts in 1988 with Especially For You.
A TV insider said: “Getting Kylie and Jason back together is a real coup and the results are hilarious.
“The episode is really tongue in cheek and they crack gags about their history and music.
“It was all kept hush-hush on set to keep the secret that they were both reuniting for the show.”
Ghosts, about a series of spirits haunting a country house, has been on screens since 2019.
The Red Nose Day special will air as part of the Comic Relief telethon on March 17.
